The Destination:
Gordian Biotechnology is a therapeutics company whose mission is to cure age-related diseases and ultimately allow people to lead longer, healthier lives. Traditional ex vivo screening methods have failed to produce effective treatments, as age-related diseases have complex causes that include interactions with the aged environment. To address this problem, Gordian’s platform is the first and only to enable pooled in vivo tests at scale in our proprietary “patient avatars”. We are able to deliver and test hundreds of therapeutics in individual animals, and scale efficacy testing to cover every therapeutic target at the beginning of the drug discovery process (more info on our website). We can choose targets that really impact disease biology, to develop and commercialize therapies for the world’s deadliest diseases, and eventually for the processes of aging itself.

About the Role:
Your primary mission as Scientist, Complex Ex Vivo Systems is to design and oversee ex vivo experiments to validate our screen results for our osteoarthritis (OA) disease program. Specifically, you will architect experimental designs, set up new assays and evaluate the safety, efficacy, and mechanism of action of potential therapeutic targets.
Your focus at Gordian is for the validation or elimination of screen hits to happen as early as possible. Gordian is unique in that our in vivo discovery delivers results at a far greater scale than even the largest pharma company. This means that we aren’t dependent on the success of a single target, but can survey the landscape of disease biology for the most promising target (or combination of targets) before moving to therapeutic development. You’ll work closely with our heads of disease biology, managing experimental schedules, and developing assays for in vivo and ex vivo workflows and setting up ex vivo systems to drive forward validation plans for our OA program.
In your first month you’ll become familiar with our technology and workflows, developing a strong understanding of the role of the preclinical team at Gordian. You’ll become proficient in our established ex vivo cultures and use your expertise to optimize and perfect the ex vivo culture systems.
By 2 months, own an entire ex vivo validation project that aims at assessing the therapeutic efficacy of 10 or more treatments across 10 or more human donor samples. . You will generate validation data in collaboration with other teams, analyze and present to stakeholders for decision-making.
At 3 months,. You’ll have established robust culture methods / delivery, generated validation data of multiple therapies. You’ll have established a medium-to-high throughput ex vivo validation process that feeds seamlessly into the in vivo validation pipeline. You’ll have independently initiated another validation project with optimized data collection, organization and analyses.
By 6 months, you’ll have completed the first round of target validation for two ex vivo systems.

You have experience in human ex vivo culture systems, specifically, with ex vivo human chondrocytes and knee joint explants (OA). You’re experienced in Cryosectioning tissues and RT-QPCR for gene expression in cell and tissue samples.
You’re experienced with ELISA, western blotting, enzymatic readouts, histology and image analysis (eg: ImageJ) and can effectively interpret the data results.
You have experience managing and collaborating on time-sensitive projects, and are comfortable guiding and collaborating across multiple teams to execute experiments at a scale beyond any single person.
Experience working in industry and working with Lentiviruses/AAVs are both pluses.
